What is Proline?
Proline is a non-essential amino acid with an unusual cyclic structure that makes it one of the most structurally distinctive of all the amino acids. Its pyrrolidine ring, which connects its side chain back to the amino group, creates a rigid kink in any polypeptide chain it occupies. This structural rigidity is not a limitation - in collagen specifically, it is a fundamental architectural requirement. Proline (alongside hydroxyproline and glycine) is one of the three amino acids that appear in the repeating Gly-X-Y tripeptide sequence that defines collagen's primary structure. Together, these three residues are understood to be essential to the formation and stability of the collagen triple helix.
In the skin, proline is present not only within the dermal collagen matrix but also as part of the natural moisturizing factor (NMF) in the stratum corneum - released there as a free amino acid during the filaggrin breakdown process. This dual presence makes proline relevant both to the deeper structural environment of the dermis and to the surface hydration network of the epidermis, distinguishing it from amino acids that participate primarily at one level.
At Sweet Chemistry, proline is formulated as part of the eleven-amino-acid complex at 1% across the three water-based formulas. Its presence alongside glycine - the other major collagen amino acid in the blend - creates a topical amino acid environment that directly addresses the most structurally significant amino acid constituents of dermal collagen simultaneously.
What does it do for skin?
In topical application, proline functions first as an NMF component - associated with water-binding within the stratum corneum's moisture-retaining network alongside PCA, sodium PCA, sodium lactate, and the other free amino acids in the Sweet Chemistry formulas. Its contribution to the NMF water-binding environment supports the same surface hydration goal as the other amino acids in the blend.
Beyond the NMF surface, proline's most studied topical relevance is its association with the collagen formation environment. Fibroblasts require proline as a substrate for new collagen synthesis - specifically for the production of both proline and its hydroxylated derivative, hydroxyproline, which stabilizes the collagen triple helix structure. When formulated alongside Matrikynes® peptides (which are associated in research with fibroblast signaling and the appearance of improved collagen density) and THD ascorbate (the vitamin C cofactor studied in connection with collagen cross-linking), topical proline contributes the raw material that the collagen biosynthesis environment is understood to require.
