What is Histidine?

Histidine is an essential amino acid, meaning the human body must obtain it from external sources as it cannot synthesize it in sufficient quantities independently. It is one of the more biochemically versatile amino acids, distinguished by its imidazole side chain - a heterocyclic ring structure that gives histidine unique metal-chelating properties and a distinctive reactivity profile compared to other amino acids. In the skin, histidine is present both as a free amino acid within the natural moisturizing factor (NMF) and as a structural component of proteins including filaggrin - the precursor protein that breaks down in the epidermis to release NMF components including histidine and PCA into the stratum corneum.

Research on histidine in the context of skin science has explored it from multiple angles. Its imidazole side chain has been studied in connection with metal ion chelation - specifically its affinity for copper and zinc, which has implications for both antioxidant support and the enzymatic environment of the skin's surface. In this context, histidine has been researched as a possible supporter of the activity of copper-dependent enzymes associated with collagen cross-linking and skin renewal processes.
